ABSTRACT

Abstract The present study has been carried out with the seed extracts of Nyctanthes arbor-tristis L. (Parijat) and evaluates its antioxidant potential and profiling the phytochemical constituents by Gas Chromatography-Mass Spectrometry (GC-MS) analysis. The antioxidant potential of the seed extracts was measured by four different in vitro assay like 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, superoxide anion free radical scavenging activity, ferric reducing antioxidant power (FRAP) and lipid peroxidation inhibition potential (LPIP) assay. The total phenol content (TPC) and total flavonoid content (TFC) were estimated. The ethyl acetate extract (EAE) of seeds showed potential DPPH free radical scavenging activity (EC50 129.49±3.55µg/ml), superoxide anion radical (EC50 969.94±8.03µg/ml) and LPIP (EC50 452.43±5.07 µg/ml) activities. The total phenol content was maximum in aqueous extract (AQE) which was 201.00±0.20 µg/mg gallic acid equivalent. The EAE was rich with total flavonoid and it was found to be 34.50±0.40 µg/mg rutin equivalent. The EAE was subjected for phytochemical-profiling using GC-MS system. The presence of different phytoconstituents supports the medicinal value of the seeds. The results suggest that EAE constitutes a promising new source of novel compounds. Further, it can be used for isolation and purification of specific compounds which have good antioxidant activities and possess useful biological activities.

ABSTRACT

This study evaluated the fatty acid composition and qualitative characteristics of meat from lambs feed in feedlot with increasing levels of canola grain. Meat obtained from 27 lambs fed 0, 8 and 16% inclusion of canola grain were analyzed. There was a reduction in the content of saturated fatty acids (SFA): lauric, heptadecanoic, and stearic; and unsaturated fatty acids (UFA): palmitoleic, oleic, and eicosatrienoic, with the increasing levels of canola grain. There was a linear reduction for the ω3 acid and the ω3: ω6 ratio, while the ω6: ω3 ratio increased. But the values observed for this ratio (ω6: ω3) were lower than 4, which is considered a satisfactory value. There was a slight increase (~2%) for the thrombogenicity index and atherogenicity index with the inclusion of canola grain. There was a reduction in the hypocholesterolemic: hypercholesterolemic ratio, with mean values of 2.09 for the diet with 0% inclusion, and 2.06 for 8 and 16% inclusion of canola. Although the inclusion of canola grain for lambs decreased the content of some UFA, reflecting alterations in the correlated nutritional properties, there is a reduction of SFA. The results for instrumental analysis, proximate composition, and sensory acceptance of the lamb meat were similar among the treatments.(AU)


Objetivou-se avaliar a composição dos ácidos graxos e as características qualitativas da carne de cordeiros terminados com níveis crescentes de canola grão na dieta. Foram estudadas amostras de carne provenientes de 27 cordeiros, confinados por 45 dias e alimentados com 00, 08 e 16% de inclusão de canola grão. Houve efeito para os ácidos graxos saturados (AGS): laurico, heptadecanoico e esteárico; ácidos graxos monoinsaturados (AGMI): palmitoleico e oleico; e ácidos graxos polinsaturados (AGPI): eicosatrienoico. Estes ácidos reduziram à medida que os níveis da canola grão aumentaram. Para os AGMI, houve efeito com maior concentração no músculo dos cordeiros alimentados sem a canola grão (49,80%). Houve uma redução linear para o ácido ω3 e para a relação ω3:ω6, enquanto que a relação ω6:ω3 aumentou. A canola grão influenciou o índice de aterogenicidade, com média de 0,57 para o tratamento com 0% de inclusão e 0,58 para os tratamentos com 8% e 16% de inclusão. Observou-se aumento linear para o índice de trombogenicidade com a inclusão da canola grão, cujas médias foram, respectivamente, 0,16, 0,17 e 0,18 para os tratamentos 0%, 8% e 16% de inclusão. Houve redução para a relação hipocolesterolêmicos-hipercolesterolêmicos, com médias de 2,09 para a dieta com 0% de inclusão, e 2,06 para 8% e 16% de inclusão da canola em grão na dieta. A canola grão não influenciou a análise instrumental, composição centesimal e atributos sensoriais da carne.(AU)

ABSTRACT Polyunsaturated fatty acids oxidize easily due to their chemical structure, causing a reduction of their nutritional properties. Nanostructured systems may be an alternative to protect fatty acids against oxidation, improving solubility and stability. Consequently, nutritional value of food is maintained as well as the sensory characteristics (color, flavor, texture, and aroma) when fatty acids are added to food products. The present study is a narrative review to introduce the potential benefits of omega-3 unsaturated fatty acids nanoparticles incorporated in food products. The literature review includes publications in English and Portuguese issued between March 1985 and March 2019, in PubMed, ScienceDirect and Web of Science databases. Manual searches were conducted in the articles references lists of the articles included to identify other relevant studies. There were studies that evaluated the stability of fatty acids in food products such as bread, fruit juice, milk, yogurt, and meat. In this study, the most used nanostructured systems for the incorporation of fatty acids were the nanocapsules and the nanoliposomes. Currently, the nanostructured system demonstrates a potential to improve protection of polyunsaturated fatty acids against oxidization and thermal degradation. In this way, they maintain their functional properties and their bioavailability increases and therapeutic efficacy and sensory properties are improved. There are several methodologies being tested, which makes it difficult to identify the most efficient formulation to protect fatty acids. Nanostructured systems seem to be the best alternative to protect polyunsatured fatty acids from oxidization. The encapsulation efficiency, particle's size and type are relevant factors to be considered to evaluate oxidization. In conclusion, the review showed that currently it is impossible to determine the most efficient methodology. Besides, nanoformulations should follow international guidelines to present more standardized and therefore more efficient particles.


RESUMO Os ácidos graxos poli-insaturados são facilmente oxidados devido à sua estrutura química, o que acarreta a diminuição de suas propriedades nutricionais. Nesse contexto, o sistema nanoestruturado pode ser uma alternativa para protegê-los contra a oxidação, melhorando a solubilidade e estabilidade. Consequentemente, quando são adicionados a produtos alimentares, o valor nutricional é mantido, bem como as características sensoriais (cor, sabor, textura e aroma). O presente estudo é uma revisão narrativa para apresentar os potenciais benefícios das nanopartículas com ácidos graxos insaturados da classe ômega-3 incorporados a produtos alimentícios. A literatura consultada incluiu publicações em inglês e em português, considerando o período entre março de 1985 e março de 2019, utilizando as bases de dados PubMed, ScienceDirect e Web of Science. Foram realizadas buscas manuais nas referências dos artigos incluídos, a fim de identificar outros estudos relevantes. Foram encontradas pesquisas que avaliaram a estabilidade dos ácidos graxos em produtos alimentícios, como pães, suco de fruta, leite, iogurte e carne. Neste estudo, as nanoestruturas mais utilizadas para a incorporação dos ácidos graxos foram as nanocápsulas e os nanolipossomas. Atualmente, o sistema nanoestruturado demonstra um potencial para melhorar a proteção desses ácidos poli-insaturados contra a oxidação e degradação térmica. Dessa forma, eles mantêm suas propriedades funcionais, aumenta-se sua biodisponibilidade e há melhora da eficácia terapêutica e das propriedades sensoriais. Existem diversas metodologias sendo testadas, o que dificulta a identificação de qual seria a formulação mais eficiente nessa proteção, mas os sistemas nanoestruturados parecem ser a melhor alternativa para proteger os ácidos graxos insaturados da oxidação. A eficiência de encapsulação bem como o tamanho e o tipo de partícula são fatores importantes a considerar na avaliação da oxidação. Em conclusão, a revisão demonstrou que atualmente a metodologia mais eficiente não é possível de ser identificada. Ademais, as nanoformulações devem seguir diretrizes do protocolo internacional para apresentar partículas mais padronizadas e, assim, eficientes.

ABSTRACT The need to obtain nutritious foods from new sources and lower waste in industry has created a high interest in studying different parts of plants or foods that today are considered waste, but could be considered by-products with high nutritional value with potential use in human diets. Pumpkin seeds are commonly considered as waste but they have a high content of fatty and amino acids, which when used as a by-product or ingredient can add value to food products. The aim of this work was to perform a wide review of the nutritional and functional properties of Cucurbita maxima seeds and their potential medicinal influence.


RESUMEN La necesidad de obtener alimentos nutritivos de nuevas fuentes y menores desperdicios en la industria ha generado un gran interés en el estudio de diferentes partes de plantas o alimentos que hoy en día se consideran desechos, pero que podrían considerarse subproductos con alto valor nutricional y uso potencial en alimentación humana. Las semillas de calabaza se consideran comúnmente como desechos, pero tienen un alto e importante contenido de ácidos grasos y aminoácidos, que cuando se utilizan como subproducto o ingrediente pueden aportar un alto valor agregado a los productos alimenticios. El objetivo de este trabajo es realizar una amplia revisión de las propiedades nutricionales y funcionales de las semillas de Cucurbita maxima y su potencial influencia medicinal asociada a ellas.

Objective To learn the latest progress in the research of ω-3 unsaturated fatty acids and provide references for the related investigator through visualized analysis of the research of ω-3 unsaturated fatty acids published in our country.Methods China Biology Medicine disc was searched by computer from the beginning until December 31,2017.Bibliographic Item Co-occurrence Mining System (BICOMS) was used to extract and summarize the data of age,author,organization,province and key words and to produce a co-occurrence matrix.NetDraw of Ucinet 6.0 was employed to draw the social network diagram of the author,organization,province and key words.And cluster analysis of the key words was performed by gCLUTO 2.0.Results A total of 1 165 studies involviug 30 provinces and cities,854 research units,3 789 authors and 1 016 significative key words were included.The cooperation of author,organization and province needed to be further strengthened.The study focused on 3 aspects:the effects of unsaturated fatty acids on apoptosis/lipid in mice/rats with related genes of diabetes/cardiovascular disease/coronary artery disease;meta-analysis of immunonutrition to improve tumor/inflammation/sepsis;effect of fish oil fat emulsion on interleukin/C reactive protein in pneumonia/lung injury.Conclusions The research of ω-3 unsaturated fatty acids develops rapidly in China,but it is mainly concentrated in a few centers of cooperation agencies.There is little cooperation among different provinces,cities,areas,and organizations and the research topics needs to be further expanded.

ABSTRACT

Objective To observe the effects of 3 neuroprotective measures on the expressions of apoptosis-related factors and their ligands (Fas and FasL) in brain tissue of neonatal rats with hypoxic ischemic brain injury. Methods One hundred and twenty Wistar rats 7 days old were selected as experimental subjects, the rats were divided into four groups: neural stem cell, erythropoietin (EPO), ω-3 unsaturated fatty acid treatment groups and hypoxic ischemic brain damage model group according to random number table method, with 30 rats in each group. Neural stem cell group, EPO group and ω-3 unsaturated fatty acid group were respectively injected with neural stem cells, EPO and ω-3 unsaturated fatty acid, each 5 mL via tail vein after modeling; the hypoxic ischemic brain damage model group was given equal volume of normal saline. At 6, 12, 24, 48 and 72 hours after administration of drug, 6 rats were sacrificed in each group, brain tissue was taken, the mRNA expression levels of Fas/FasL, protein expression levels of Toll-like receptor 4 (TLR4), nuclear transcription factor-κB (NF-κB), tumor necrosis factor-α (TNF-α), interleukin (IL-1β, IL-6) and cell apoptotic rate in hippocampus tissue were measured. Results ① mRNA expressions: the mRNA expressions of Fas and FasL of the 3 experimental groups were significantly lower than those of the hypoxic ischemic brain damage model group, the degrees of descent after administration for 24 hours were the most significant, neural stem cell treatment group < EPO treatment group < ω-3 unsaturated fatty acid treatment group < hypoxic ischemic brain damage model group [Fas mRNA expression (2-ΔΔCt): 140.5±2.9, 156.4±2.5, 165.2±2.7 vs. 173.7±2.8, FasL mRNA expression (2-ΔΔCt): 143.1±4.3, 154.6±1.5, 160.7±1.4 vs. 174.7±2.8], the differences were statistically significant (all P < 0.05). ② Protein expressions: the protein expressions of TLR4, NF-κB, TNF-α, IL-1β, IL-6 of the 3 experimental groups were significantly lower than those of the hypoxic ischemic brain damage model group (TLR4/GAPDH: 0.7±0.2, 0.6±0.1, 0.2±0.1 vs. 1.4±0.1; NF-κB/GAPDH: 6.7±0.4, 5.3±0.1, 1.1±0.2 vs. 11.2±0.3; TNF-α/GAPDH: 14.3±1.4, 11.2±1.2, 3.2±2.1 vs. 23.2±0.5; IL-1β/GAPDH: 9.4±0.2, 7.4±0.3, 2.2±0.3 vs. 13.4±0.1; IL-6/GAPDH: 36.2±4.4, 39.3±1.5, 26.2±2.1 vs. 51.4±1.4, all P < 0.05), the protein expression levels of above indexes in neural stem cell treatment group < those of EPO treatment group < those of ω-3 unsaturated fatty acid treatment group < those of hypoxic ischemic brain damage model group. ③ Apoptotic rates:after drug administration, the apoptotic rates of the ω-3 unsaturated fatty acid group, EPO treatment group, neural stem cell treatment group were obviously lower than the rate of model group [(3.7±0.3)%, (3.4±0.2)%, (2.5±0.1)% vs. (5.5±0.4)%, all P < 0.05]. Conclusion The mRNA expressions of Fas/FasL in the brain of neonatal rats with hypoxic-ischemic brain damage are high, and the treatment with each of the following agents; neural stem cells, EPO and ω-3 unsaturated fatty acid can reduce the mRNA expressions of Fas/FasL in such rats' brain tissues.

ABSTRACT

Background: Breast cancer is the second cause of death in women in developed and undeveloped countries, including Colombia. A high percentage of these tumors is estrogen dependent, for which the hormonal treatment is the most used therapy in breast cancer. Currently, the first line treatment for breast tumor in postmenopausal women is the letrozole, an aromatase enzyme inhibitor that avoids the transformation of androgens to estrogens. Since letrozole produced adverse effects on patients, there is a requirement for new alternative treatments. Furthermore, omega fatty acids (ω-FA), essential as they are obtained from the normal diet or from dietary supplements, have demonstrated nutraceutical potential because of their anti-inflammatory or pro-inflammatory activity. Nonetheless, there is controversy in in vitro, in vivo and epidemiologic reports regarding their preventive or inducing activities of carcinogenesis in animals and humans, depending on the structure of the ω-FA. Objectives: This review aims to show the main in vitro, in vivo and epidemiologic evidences of the chemotherapeutic potential of ω-3 and ω-6 FA in different types of neoplasm, particularly in breast cancer, in individual or combined treatments with diverse antineoplastics. Methods: PubMed and Science Direct databases revealed the most representative studies, published during the last two decades, about ω-3 and ω-6 FA, breast cancer and the principal therapeutic strategies for this neoplasm. Findings were presented in separated topics to provide an overview of ω-FA and their potential in treatments for breast cancer. Results: Patients treated with estrogens and progesterone derivate have shown predisposition to develop breast cancer after two years of continued therapy. Furthermore, ω-FA with known nutraceutical potential have demonstrated their potential as adjuvants in the treatment against different neoplasms, like hepatic and colon cancer. Conclusions: Current therapies for breast cancer and their low efficacy in the long term led to explore new alternative treatments with ω-FA. These essential fatty acids in daily consumption could enhance the antineoplastic agent effect. Nevertheless, metabolism of the ω-FA must be considered for this use.


Antecedentes: el cáncer de mama es la segunda causa de muerte de mujeres en países desarrollados y no desarrollados, incluido Colombia. La mayoría de estos tumores son dependientes de estrógeno por esa razón, la terapia más utilizada es la hormonal. Actualmente, el tratamiento de primera línea en mujeres posmenopáusicas es el letrozol, inhibidor de la enzima aromatasa, que evita la conversión de andrógenos en estrógenos. El letrozol causa efectos adversos en las pacientes, lo cual motiva la búsqueda de nuevas alternativas que disminuyan estos efectos. Los ácidos grasos omega, esenciales en la dieta regular o suplementaria, han mostrado su potencial nutracéutico ambivalente, como antiinflamatorios o proinflamatorios. Debido a esto, existe controversia en distintos reportes a nivel in vitro, in vivo y epidemiológicos sobre la actividad preventiva o quimioterapéutica de los ω-3 y ω-6 AGOs. Objetivos: el aporte de este artículo, es mostrar las principales evidencias in vitro, in vivo y epidemiológicas del potencial quimioterapéutico de los AGOs en tratamientos individuales y combinados con antineoplásicos, en distintos tipos de cánceres, particularmente en el cáncer de mama. Métodos: se revisaron las bases de datos PubMed y Science Direct y se seleccionaron los estudios más representativos de las dos últimas décadas sobre ω-3 y ω-6 AGOs y las principales estrategias usadas en el cáncer de mama. Los hallazgos se presentan en temas separados, primero una visión general de los AGOs y luego su potencial bioactivo en tratamientos contra el cáncer de mama. Resultados: la mayoría de los estudios en pacientes con cáncer de mama, tratadas con estrógenos y derivados de progesterona, han mostrado predisposición a desarrollar cáncer de mama después de dos años de terapia continua. De otro lado, los AGOs han demostrado su potencial como adyuvantes en el tratamiento en diferentes cánceres como el de colon y hepático. Conclusiones: las terapias actuales para el cáncer de mama y su baja eficacia a largo plazo exigen explorar nuevas alternativas de terapias, que incluyen los AGOs podrían potenciar fármacos, no obstante, es necesario tener en cuenta, el metabolismo de los AGOs, para uso

Resumen Objetivo. Identificar y evaluar la capacidad antimicrobiana de los metabolitos aislados a partir del proceso de fermentación en una cepa de Mucor circinelloides. Método. En el presente estudio se trabajó la cepa nativa de Mucor circinelloides SPG 321 suministrada por el Grupo de Investigación de Biotrasformación (GIBUJ), con el fin de evaluar su actividad antimicrobiana. La cinética de crecimiento determinó que la ideofase culminó a la hora 264, hora determinada como la finalización de la fermentación. Se separó el micelio del caldo y posteriormente se fraccionaron los extractos con cromatografías de capa fina y columna en sílica gel, eluidas con diferentes relaciones de solventes. Resultados. Los resultados arrojados por la técnica de gases acoplado a masas CG-EM confirman la importancia de Mucor circinelloides en la producción de ácidos grasos insaturados. A partir del micelio se obtuvo un esterol, compuesto M. cB3. La fracción CHCl3 en biomasa mostró actividad inhibitoria para los microorganismos Gram positivos.


Abstract Objective. To identify and evaluate the antimicrobial capacity of the metabolites isolated from the fermentation process in a strain of Mucor circinelloides. Method. For this study it was taken the 321 SPG native strain of Mucor circinelloides provided by GIBUJ. The kinetics growth was made, observing the ending of the idiofase at hour 264, hour taken for the conclusion of fermentation. Mycelium was separated from the broth and then purified with thin layer chromatography and silica gel column, eluted with different relations of solvents. Results. The results given by the technique of coupled gases with GC-MS mass confirm the importance of Mucor circinelloides in the production of unsaturated fatty acids. A sterol was obtained from the mycelium, M.cB3 compound. The fraction CHCl3 in biomass showed inhibitory activity for Gram-positive microorganisms.

Objective To investigate the effects of enteral immunonutrition supplemented with omega-3 polyunsaturated fatty acid (ω-3 PUFA) on inflammatory response,intestinal mucosal barrier function and the prognosis in patients with severe traumatic brain injury (sTBI).Methods 122 patients of sTBI hospitalized between January 2015 and December 2016 were randomly divided into experimental group (ω-3 PUFA,n=61) and control group (n =61).The serum levels of tumor necrosis factor-α (TNF-α),interleukin (IL)-6 and neuron specific enolase (NSE) were tested with enzyme linked immunosorbent assay.Meanwhile,D-lactate acid and intestinal fat acid binding protein (I-FABP) were evaluated by enzymology spectrophotometer method.After 14 days of treatment,the Glasgow Coma Scale (GCS) scores,Acute Physiology and Chronic Health Evaluation (APACHE) Ⅱ scores and prognoses of both groups were compared.Results The serum levels of inflammatory factors (TNF-α and IL-6),intestinal mucosal barrier function indicators (D-lactate acid and I-FABP) and NSE proteins significantly increased after sTBI (P =0.01).Compared with the control group,the experimental group on day 3 had significantly lower serum levels of inflammatory factors [TNF-α:(107.77± 19.79) μg/Lvs.(151.76±21.65) μg/L,P=0.01;IL-6:(76.85±7.15) μg/Lvs.(105.27±10.12) μg/L,P=0.01] and intestinal mucosal barrier function indicators [D-lactate:(69.81 ±6.32) μg/L vs.(89.80± 8.75) μg/L,P=0.03;I-FABP:(40.81±6.73) μg/Lvs.(56.60±8.58) μg/L,P=0.01].On day 7,the experimental group had significantly lower expression of NSE proteins than the control group [(13.63± 2.53) μg/L vs.(19.12±3.00) μg/L,P=0.02].The experimental group received better prognosis compared to the control group on day 14 [GCS scores:(9.74±0.76) vs.(8.44±0.53),P=0.04;APACHE Ⅱ scores:(14.67±1.37) vs.(17.53±1.47),P=0.03].The experimental group also had fewer days in hospitalization [(19.37±2.27) d vs.(25.42±2.61) d,P=0.01].Conclusion Enteral immunonutrition supplemented with ω-3 PUFA can effectively regulate the inflammatory response,and reduce impairment to the intestinal mucosal barrier function and damage to neurons in patients with sTBI.

Fish oil is an important nutrient component in rainbow trout bone, and the optimization of extraction by enzymatic hydrolytic method is of great significance.This study selected the alkaline protease as the hydrolytic enzyme, and optimized the process conditions of enzymatic hydrolysis of rainbow trout fish using single factor analysis method.Effects of several factors on the extraction of fish oil were studied, including the ratio of material to liquid, pH, enzymatic hydrolysis time, enzymatic hydrolysis temperature and amount of enzyme.Fatty acids were identified by gas chromatography-mass spectrometry (GC-MS).Results showed that the optimum extraction parameters of enzymatic hydrolysis were as follows: 2000 U/g alkaline protease, ratio of material to liquid of 1∶1 (w/w), pH 7.5, and extraction at 55℃ for 3h.It was found that the main composition of rainbow trout bone oil was unsaturated fatty acid with the content of 80.4% (w/w).The relative content of monounsaturated fatty acid and polyunsaturated fatty acid was about 76.9% (w/w) and 23.1% (w/w), respectively.The total content of EPA and DHA was 3.4% (w/w).This study optimized the extraction method of rainbow trout fish oil, analyzed and identified the main volatile compounds, and identified the main substances contributing to fish oil flavor.The method thus was of significance for the analysis and identification of fish oil products.

O objetivo deste trabalho foi avaliar os efeitos da adição de fontes de lipídeos naturais e protegidos da degradação ruminal na dieta de bovinos Nelore confinados sobre características da carne, concentrações de lipoproteínas sanguíneas e perfil de ácidos graxos da gordura do músculo Longissimus dorsi. Foram utilizados 120 bovinos Nelore, não castrados (366,9±28,7kg), com aproximadamente 24 meses. O delineamento experimental foi inteiramente casualizado, com três tratamentos: (CONTR) sem fonte adicional de lipídeo, (GDESP) com fonte de lipídeo natural (torta de algodão) e (GPROT) com fonte de lipídeo protegido rico em ácidos graxos poli-insaturados. Foram utilizadas 24 baias, cinco animais/baia, com oito repetições/tratamento, sendo as baias consideradas unidades experimentais. Foi observada (P<0,05) interação entre tratamento e dias de mensuração para colesterol e LDL, com maiores valores do tratamento GPROT nos dias 1 e 15, tendo o dia 84 valor intermediário aos demais tratamentos. Observaram-se efeitos de dias de mensuração, em que VLDL e triglicerídeos apresentaram valores crescentes nos dias 15, 84 e 1; enquanto o HDL teve maior valor no dia 1. Foi observado efeito (P<0,05) de tratamento para o perfil de ácidos graxos, com diminuição do C14:1, C16:1 e C17:1 e aumento do ácido transvaccênico, C18:2, ácidos graxos poli-insaturados (AGPI) e da relação AGPI: AGMI (ácidos graxos monoinsaturados) para os tratamentos GPROT e GDESP. Adição de lipídios na dieta, independentemente da fonte, promove melhora na composição de ácidos graxos da carne de bovinos Nelore confinados, aumentando a quantidade de ácidos graxos insaturados sem alterar as características qualitativas da carne.


The objective of this work was to evaluate the effects of adding sources of natural lipids and rumen-protected lipid source rich in polyunsaturated fatty acids to the diet of feedlot Nellore cattle on meat characteristics, blood lipoprotein concentration and fatty acid profile of subcutaneous fat of Longissimus dorsi. One hundred and twenty 24-mo-old Nellore yearling bulls were used (366.9±28.7kg). The experimental design was completely randomized, replicated 8 times (5 bulls/pen), with pens being considered experimental units and consisting of three treatments: (CONTR) without additional source of lipid, (GDESP) with source of natural lipid (cottonseed cake), and (GPROT) with rumen-protected lipid source rich in polyunsaturated fatty acids. Interaction (P<0.05) was observed between treatment and day of measurement for cholesterol and LDL, with values greater GPROT treatment on days 1 and 15, and day 84 intermediate value to the other treatments. Effects (P<0.05) were observed on days of measurement, which showed an increase in VLDL and triglyceride values on days 15, 84 and 1; while HDL had higher values on day 1. Effect (P<0.05) of treatment for the fatty acid profile showed decreased C14:1, C16:1 and C17:1 and increased transvaccênico acid, C18:2, polyunsaturated fatty acids (PUFA) and the ratio PUFA:MUFA (monounsaturated fatty acids) for GPROT and GDESP treatments. Additional fat in the diet, regardless of the source, promotes improvement in fatty acid composition of meat from feedlot Nellore cattle, increasing the amount of unsaturated fatty acids without changing the qualitative characteristics of meat.

Introdução: Mudanças na alimentação e atividade física das populações elevaram a incidência de doenças crônicas não-transmissíveis associadas à adiposidade corporal. Este quadro contribui para mortalidade cardiovascular, motivando iniciativas em saúde pública visando à prevenção. Há evidências de que populações que consomem a dieta mediterrânea apresentam menor mortalidade por todas as causas, inclusive cardiovasculares. Os benefícios desta dieta, rica em fibras, gorduras insaturadas e polifenóis, parecem decorrer da atenuação da inflamação, envolvida na gênese de doenças cardiometabólicas. Objetivo: Este estudo investigou os efeitos da modificação de uma refeição diária, o desjejum, de forma a incluir alimentos mediterrâneos, sobre o metabolismo lipídico, glicídico, inflamação subclínica e expressão de genes inflamatórios. Métodos: Foi um ensaio clínico cruzado com duração total de 10 semanas, incluindo 80 adultos com excesso de peso, não-diabéticos. Os participantes passaram por 2 intervenções de 4 semanas no desjejum, com wash-out de 2 semanas entre elas. Os desjejuns, brasileiro e modificado, foram isocalóricos, diferindo quanto ao conteúdo de fibras e tipos de ácidos graxos. Antes e após cada intervenção foi realizado teste de sobrecarga de gorduras (FTT) com refeição rica em gorduras (saturadas e insaturadas MUFA e PUFA, dependendo da intervenção) e coletas sanguíneas seriadas até 240 minutos para determinação de glicose, insulina, lípides e marcadores inflamatórios.


Introduction: Changes in dietary pattern and physical activity of populations have elevated the incidence of chronic non-communicable diseases associated with increased adiposity. Evidence has shown that populations consuming Mediterranean diets have lower mortality from all causes, including cardiovascular diseases. The benefits of this diet rich in fiber and unsaturated fats, derived in part on the effects of these nutrients on inflammatory condition that triggers cardiometabolic diseases. Objective: This study investigated the effects of changing a meal of Brazilian menu, the breakfast, in order to approximate it to the Mediterranean pattern on lipid and glucose metabolism, subclinical inflammation and also on the expression of inflammatory genes. Methods: This study was a crossover trial lasting a total of 10 weeks, including 80 overweight adults, nondiabetic without drug treatment for dyslipidemia. Participants who met the inclusion criteria underwent two 4-week interventions in breakfast, with wash-out of two weeks between them. The breakfasts (Brazilian and modified) were isocaloric, differing according to fiber and types of fatty acids contents. Before and after each intervention, fat tolerance tests with meals rich in fat (saturated and unsaturated depending on the intervention) were perfomed, with blood sample collections for glucose, insulin, lipids and inflammatory markers up to 240 minutes. Also, expression of inflammatory genes before and after each intervention was analyzed.

ABSTRACT

This study was aimed to analyze the plasma metabolicomics pathway in rats with heart blood stasis syn-drome. KEGG database was used in the signal pathway analysis. HMDB was used in the analysis of molecular metabolite annotation, enzyme or transporter associated and its related properties. The metPA network software was used in the visualization of metabolite path. The results showed that 9 metabolites involved in 15 metabolic pathways. Among them, the P-value of metabolic pathway of pantothenate and CoA biosynthesis, propanoate metabolism, biosynthesis of unsaturated fatty acids was less than 0.05. It was concluded that the metabolic pathways of pan-tothenate and CoA biosynthesis, propanoate metabolism, biosynthesis of unsaturated fatty acids were involved with the pathological process of rats with heart blood stasis syndrome.

ABSTRACT

Nuts are a highly nutritious food that contains many useful healthy unsaturated fatty acids, vitamins and minerals. It is clear that nuts have a beneficial impact on many cardiovascular risk factors. However, little epidemiological evidence exists regarding the effects of nuts on diabetes risk. Therefore, in order to understand the role of nuts in prevention and management of diabetes, more long-term clinical studies are needed to clarify the possible effects of nuts on diabetes. Nuts have high energy and fat content, and thus have a healthy dietary component. For this reason, regular nut consumption can be recommended to avoid excessive caloric intake above normal requirements.

ABSTRACT

Objective To investigate the effect of intestinal lymphatic duct ligation and ω-3 polyun saturated fatty acids on intestinal and distant organ in intestinal ischemia-reperfusion injury. Methods Totally 40Sprague-Dawley (SD) male rats (SPF grade)after gastrostomy were equally randomized into sham group (Sham), enteral nutrition (EN) group, enteral nutrition and lymphatic duct ligation (EN + L) group, ω-3 polyunsaturated fatty acids (ω-3PUFA) group, and ω-3PUFA and lymphatic duct ligation (ω-3PUFA + L) group. After 7 days of nutritional intervention, rats were subjected to 60 minutes of intestinal ischemia, ischemia plus mesenteric lymph duct ligation, or sham procedures. After 3 days of continuous nutrition intervention using the original nutrient, lymph nodes, lung, intestine, liver, and blood specimens were harvested. Intestinal permeability and morphology, results of bacterial cultures, and serum cytokines were observed or detected. Result After 3 days of intestinal ischemia-reperfusion (I/R), the body weights of rats in EN group significantly decreased when com pared with the pre-I/R levels (P < 0.05), while the body weights of rats in EN + L group were significantly lower than those in ω-PUFA group and ω-PUFA + L group (P < 0. 05). After one day of intestinal ischemia-reperfusion (I/R), the L/M significantly increased in each group (P <0.05 or P <0. 01). After 3 days of intestinal ischemiareperfusion (I/R) , the L/M were significantly lower than the level one day after ischemia- reperfusion in EN + L group, ω-PUFA group, and ω-PUFA + L group (P < 0.05). The L/M in EN group and EN + L group were significantly higher than that in ω-PUFA + L group (P < 0. 05). The mucosa thickness and villus height of jejunum in ω-PUFA group and ω-PUFA + L group were significantly higher than those in Sham group, EN group, and EN + L group (P < 0. 01 or P < 0. 05). The mucosa thickness and villus height of ileum in ω-PUFA group and ω-PUFA +L group were also significantly higher than those in EN group (P < 0.05). In ω-PUFA + L group, the serum endotoxin level and tumor necrosis factor-α level were significantly lower than those in EN group (P < 0.05), interleukin (IL) -6 level was significantly lower than that in the ω-PUFA group (P < 0.05), and IL-1 β level was significantly lower than those in other groups (P < 0. 05). In EN group, the lung cell apoptosis index was significantly higher than those in other groups (P < 0.05)and the levels of inducible nitric oxide synthase (iNOS)and myeloperoxidase (MPO) were significantly higher than those in ω-PUFA + L group (P < 0. 05). The level of iNOS was also significantly higher in EN + L group than that in ω-PUFA + L group (P < 0.05). Conclusions Sixty minutes of intestinal ischemia can cause intestinal injury, intestinal barrier dysfunction, and increased permeability of intestine. After 72 h of reperfusion, the intestinal injury can be partially recovered and the permeability can be lower than the post-ischemia level; however, bacterial endotoxin translocation and lung apoptotic cells still exist. Intestinal lymphatic ligation can alleviate the lung damage, promote repair of intestinal mucosa, reduce endotoxin translocation, and attenuate the systemic inflammatory response. EN added with ω-3PUFA is remarkably superior to conventional EN.

ABSTRACT

The effects of changes in fatty acid composition of the cell membrane on different biological functions of Salmonella typhimurium have been studied with the help of a temperature sensitive fatty acid auxotroph which cannot synthesise unsaturated fatty acids at high temperature. On being shifted to nonpermissive temperature the cells continue growing for another one and half to two generations. The rates of protein and DNA syntheses run parallel to the growth rate but the rate of RNA synthesis is reduced. Further, there is a gradual reduction in the rate of transport of exogenous uridine and thymidine into the soluble pool. The transport process can be restored by supplementing the growth medium with cis-unsaturated fatty acids but not trans-unsaturated ones although the growth of the cells is resumed by supplementation with either cis or trans-unsaturated fatty acids. However, supplementation with trans, trans-unsaturated fatty acids leads to only partial recovery of the transport process. The rate of oxygen uptake is also affected in cells grown in the presence of the trans-unsaturated fatty acids, elaidic acid and palmitelaidic acid. Analysis of cells grown under different fatty acid supplementation indicate that fatty acid composition of the cell membrane, especially the ratio of unsaturated to saturated fatty acids varies with temperature shift and supplementation of the growth media with fatty acids.
